Clinical CDK4/6 inhibitors induce selective and immediate dissociation of p21 from cyclin D-CDK4 to inhibit CDK2

We developed a live cell protein binding assay to show that in addition to their catalytic function, clinical CDK4/6 inhibitors have a non-catalytic role, dramatically decreasing p21 affinity for CDK4 complexes allowing for binding and inhibition of CDK2 complexes. Interestingly, this effect on p21 affinity is specific for CDK4 but not CDK6 complexes.
